Anti-Peri-Implantitis Effect Of Modified Ti In Hyperlipidemic Microenvironment

The present study aims to develop dual-functional implants to combat peri-implantitis in hyperlipidemic microenvironment and test pathogenic bacteria inhibition and osteogenesis inducing effects of modified implants. Oxygen non-thermal plasma (P) treated Titanium (Ti) was grafted with organosilicon quaternary ammonium salt, yielding Ti-P-TMTSPA (Ti-PTA). Physicochemical properties and biocompatibility were evaluated. In hyperlipidemic microenvironment, effects of Ti-PTA on proliferation and osteogenesis inducing of osteoblasts were explored. Antibacterial capacity of Ti-PTA against Porphyromonas gingivalis (P. gingivalis) and Fusobacterium nucleatum (F. nucleatum) was also assessed. A model of dental implanted rats with hyperlipemia was established to test in vivo osteogenesis inducing and antibacterial ability. Ti-PTA was synthesized. The modification did not affect original morphology and roughness (p>0.05), which meanwhile exhibited improved hydrophilicity (p<0.05) and favorable biocompatibility (p>0.05). Ti-PTA could reverse growth inhibition effect hyperlipidemic microenvironment brought and boost the proliferation of osteoblasts to 2 folds of Ti (p<0.05). At the same time, ALP activity and mineralization level of osteoblasts on Ti-PTA was 4 folds of Ti (p<0.05). Meanwhile, Ti-PTA reduced the metabolism level of P. gingivalis and F. nucleatum to 30% of Ti (p<0.05). More dead bacteria were observed on Ti-PTA. Ti-PTA showed greater osseointegration promotion and antibacterial capacity of Ti in vivo (p<0.05). Osteogenesis inducing and antibacterial Ti-PTA was developed, which holds promising potential for prevention and treatment of peri-implantitis in hyperlipidemic microenvironment.

Abstract

Osteogenesis inducing and antibacterial Ti-PTA was developed, which holds promising potential for prevention and treatment of peri-implantitis in hyperlipidemic microenvironment.
