Trxr-Mimetic Se/S-Carbon Dots Promote Infected Wound Healing
To construct selenium-sulfur doped carbon dots (SE/S CDs) with the biomimetic activity of thioredoxin reductase, and to explore their effect on the healing of infected wounds. SE/S CDs were synthesized by the hydrothermal method, and their physical and chemical properties were characterized. Antibacterial activities in vitro and in vivo were carried out to verify their antibacterial activity. CCK8 assay, RT-PCR, DCFH-DA assay and immunofluorescence were used to verify the regulatory effects of SE/S CDs on antioxidant, anti-inflammatory and angiogenesis processes. A mouse model of infected skin defect was established, and histological staining was employed to evaluate the skin healing effect. Selenium-sulfur doped carbon dots with the biomimetic activity of thioredoxin reductase were synthesized. Antibacterial experiments showed that SE/S CDs had a significant inhibitory effect on Staphylococcus aureus and Escherichia coli. SE/S CDs could effectively scavenge reactive oxygen species, inhibit the M1 phenotype of macrophages and promote the M2 phenotype, and promote blood vessel regeneration. In vivo experiments demonstrated that, compared with the control group, the SE/S CDs group could promote wound healing more rapidly. SE/S CDs with the biomimetic activity of thioredoxin reductase achieve the healing of infected wounds by coordinately regulating the antibacterial and anti-inflammatory microenvironment and promoting angiogenesis, providing a new strategy for the repair of infected skin defects.
