In Vitro Study Of Hsa-Stabilized Acp For Dentin Remineralization
To prepare a human serum albumin-stabilized amorphous calcium phosphate (HSA-CaP) solution and investigate its efficacy in occluding dentinal tubules and promoting dentin remineralization. Human serum albumin, calcium dihydrogen phosphate, and calcium chloride were dissolved in a certain ratio, and the pH of the solution was adjusted to 7.0 to prepare the HSA-CaP solution. DLS was used to measure the particle size distribution. Cryo-TEM and SAED were utilized to observe the morphology and structure of HSA-CaP. XRD and FTIR were employed to analyze the composition of the products after HSA-CaP transformation in simulated saliva. Dentin slices were randomly divided into four groups: blank group, CPP-ACP group, sodium fluoride group, and HSA-CaP group. After incubation in simulated saliva for 7 days, SEM was used to observe the surface and cross-sectional morphology of the dentin. The HSA-CaP solution was successfully prepared. DLS measurements showed that the average particle size of HSA-CaP was approximately 7 nm. Cryo-TEM and SAED results indicated that HSA-CaP existed in the form of amorphous calcium phosphate nanoclusters. XRD and FTIR analyses confirmed that HSA-CaP transformed into HSA-hydroxyapatite in simulated saliva. SEM observations revealed that the HSA-CaP group formed a thicker mineralized layer on the dentin surface, sealing the vast majority of dentinal tubules, which was superior to the other groups. The HSA-CaP solution can effectively seal exposed dentinal tubules and has the potential for treating dentin hypersensitivity.
