O-Glcnac Modulates Nasal Inverted Papilloma Progression And Malignancy
This study investigated the role of O-GlcNAc glycosylation in nasal inverted papilloma (NIP) pathogenesis and malignant transformation, addressing its understudied mechanistic relevance (Zhao et al., 2016, Laryngoscope 126:E141-7). Thirty-five NIP patients, 30 sinonasal squamous cell carcinoma (SNSCC) patients, and 30 controls (normal mucosa) were enrolled (Ethics Committee-approved, Dalian Medical University, 2016–2017). Immunohistochemistry, RT-qPCR, and Western blotting assessed O-GlcNAc and ogt/oga expression. SCC6 and CNE-E1 cell lines were transfected with ogt/oga siRNA for functional assays (CCK-8, wound healing, Transwell). Statistical significance (P < 0.05) was determined via one-way ANOVA (SPSS 20.0). O-GlcNAc levels were elevated in NIP and SNSCC tissues versus controls (P < 0.05), peaking in SNSCC. ogt mRNA correlated positively with malignancy. siRNA-mediated OGT inhibition reduced SCC6 and CNE-E1 proliferation, migration, and invasion, while OGA knockdown enhanced these traits (P < 0.05). O-GlcNAc glycosylation drives NIP progression and malignancy, highlighting its potential as a therapeutic target for recurrence and malignant transformation. This study provides novel insights into NIP pathogenesis and clinical management.
