Effect Of Indole3carboxaldehyde On Bone Of Estrogen-Deficient Periodontitis Mice
To investigate the effect and mechanism of indole-3-carboxaldehyde on bone metabolism in estrogen-deficient periodontitis mice. Thirty-two female C57BL/6J mice were randomly divided into four groups: sham operation group, ovariectomized group, ovariectomized periodontitis group(Ovx_Lig), and ovariectomized periodontitis + indole-3-carboxaldehyde group(Ovx_Lig+ICA). After silk thread ligation, indole-3-carboxaldehyde was administered by gavage for 8 weeks. The mice were then euthanized, and the femur and maxilla were collected for micro-CT scanning. TRAP staining and immunohistochemistry were used to detect changes in bone metabolism. Colon tissues were collected for immunohistochemistry and qPCR to detect the expression of AhR in the mouse intestine. Transcriptome analysis was performed to detect changes in the transcriptional level of intestinal tissues. The alveolar bone resorption in the maxilla of the Ovx_Lig+ICA group was significantly lower than that in Ovx_Lig group and the BMD and trabecular thickness of the femur in the Ovx_Lig+ICA group were significantly higher than those in the Ovx_Lig group. The number of osteoclasts in the maxilla and femur of the Ovx_Lig+ICA group was significantly reduced and the expression of OCN and Runx2 in femur tissue increased significantly. The expression of AhR in the intestinal tissues of the Ovx_Lig+ICA group was significantly increased and the IL-17 signaling pathway was significantly downregulated in Ovx_Lig+ICA group. Indole-3-carboxaldehyde may alleviate bone resorption in estrogen-deficient periodontitis mice by improving intestinal inflammation through AhR signaling pathway.
