Impact Of PPC Scaffolds On Alveolar Bone Defect In Rats
The purpose of this study was to construct a Polycaprolactone-Polyethyleneglycol-Concentrated growth factor ( PCL-PEG-CGF, PPC) composite scaffold, and to explore the biological safety of the scaffold and its effect on the repair of alveolar bone defects in rats. The PCL-PEG-CGF composite scaffold was prepared by integrating the PCL-PEG scaffold with the CGF membrane. The scaffold was implanted in rats to detect its biosafety and degradation. The rat model of mandibular alveolar bone defect was established, and the effect of the scaffold on the repair of alveolar bone defect was investigated by Masson staining, immunohistochemistry, and micro-CT. The PCL-PEG-CGF composite scaffold has a fishnet-like microstructure and has good biosafety in vivo. The scaffold gradually degraded after 8 weeks of implantation. The scaffold up-regulated ALP expression at 2 weeks after surgery and up-regulated BMP-2 expression at 8 weeks after surgery. Micro CT showed that the scaffold could effectively promote the formation of new bone in the alveolar bone defect area. PCL-PEG-CGF scaffold has good in vivo biosafety and can promote alveolar bone regeneration, which is expected to become a new material to promote alveolar bone regeneration.
