Vorasidenib: A narrative drug review
ABSTRACT Isocitrate dehydrogenase (IDH) mutations (IDH1 and IDH2) are observed in malignancies such as low-grade gliomas, acute myeloid leukemia, cholangiocarcinoma, and chondrosarcoma. Vorasidenib is a targeted therapy for IDH-mutant gliomas that significantly delays disease progression and the need for additional anticancer intervention, with minimal side effects. For this review, we searched online databases including PubMed, Google Scholar, and websites such as the United States Food and Drug Administration, UpToDate, and Medscape using the search terms “Vorasidenib”, “IDH mutant”, “IDH mutant inhibitors”, and “low-grade glioma”. We identified 20 articles published between January 2017 and January 2025, of which 13 were included in this review. This article focuses on the indications, side effects, safety profile, drug interactions, pharmacokinetics, and pharmacodynamics of vorasidenib.
