Experimental study of a portable integrated optical scanning holographic microscope for 3D bioimaging
This study develops a compact optical scanning holographic microscopy (OSHM) system integrating high - precision components into a portable cage platform (60cm × 60cm, 22.45kg). On this basis, by leveraging the structural advantages of the integrated components, the system-which combines a high numerical aperture microscope objective with dual-spherical-wave interference illumination on the sample-achieves a lateral resolution of 588 nm. Experimental verification shows that the system can achieve high-resolution 3D imaging of biological samples and demonstrates imaging results in samples such as blood cells, plant cells, and mycelium. The introduced and validated dual-spherical-wave interference encoding mechanism overcomes the limitations of conventional plane-wave-spherical-wave configurations, providing a practical example of optical scanning holography in biomedical applications.
