Abstract B127: Unravelling the chemotherapeutic potential of taxifolin ruthenium-p-cymene complex in breast carcinoma and its repurposing in colon cancer

Abstract Colorectal cancer (CRC) is still a common malignancy with associated mortality. In the era of cancer relapse and multiple drug resistance, the combinational therapy using natural, metallic and organometallic compounds has grabbed keen attention as an alternative, even though some controversies still exist regarding its prolonged use. This research evaluated the anticancer potential of taxifolin ruthenium-p-cymene against colon cancer. The anticancer activity of the taxifolin ruthenium-p-cymene complex (TaxRu) was investigated using an in vivo method with a mouse colon cancer model. Dextran Sulphate Sodium (DSS) in drinking water were given to male C57BL/6 mice for one week to induce colon cancer, and TaxRu (200 mg/kg ) was administered for another 3 weeks to investigate its antitumor activity. The blood samples were collected, and serum was isolated to assess the levels of inflammatory cytokines by ELISA. Despite that, colons were harvested from the animals to isolate RNA to for mRNA expression by using RT-PCR and tissue homogenate was prepared to determine antioxidant activity. TaxRu was significantly reduced proinflammatory cytokines such as TNF-a, IL-1b, and IL-6 (****p < 0.0001), while upregulation of antioxidant enzymes (SOD, CAT and GPx) was found (****p < 0.0001) to be highly significant. Despite of, TaxRu significantly increased mRNA expression of epithelial markers ZO-1 and E-cadherin (***p < 0.001, ****p < 0.0001), whereas a significant downregulation of mesenchymal and fibrotic markers i.e. vimentin and TGF-β (**p < 0.01). Inclusion TaxRu repressed the angiogenesis and EMT process by decreasing the elevated levels of cytokines. The upregulation of antioxidants inhibits tumorigenesis , and the expression level of EMT biomarkers showed promising functionality for colon cancer. Further research work is needed to explore the mechanistic pathway, which will eventually help us to understand complex molecular crosstalk. Therefore, TaxRu can be considered a formidable drug for colorectal cancer. Citation Format: Barshana Bhattacharya, Abhijit Das, Souvik Roy, Pranab Kumar. Sahoo.
