State-of-the-art in palladium-catalyzed Denitrative cross-coupling reactions

Abstract The palladium-catalyzed denitrative coupling of nitroarenes has emerged as a transformative strategy in synthetic organic chemistry, offering an efficient and sustainable alternative to traditional cross-coupling methods that rely on aryl halides or pseudohalides. Nitroarenes, long regarded as terminal or unreactive functional groups, are now recognized as versatile electrophilic partners for the construction of carbon–carbon (C C) and carbon–heteroatom (C–X) bonds. This review highlights recent progress in palladium-catalyzed denitrative coupling reactions, focusing on key advances in the formation of C C, C O, C N, C H, and sulfone linkages. The success of these transformations is largely driven by the development of highly active Pd catalyst systems supported by tailored ligands—particularly electron-rich phosphines and N-heterocyclic carbenes—that facilitate oxidative addition into the challenging C–NO₂ bond. Mechanistic insights, including oxidative addition pathways, transmetalation, and reductive elimination steps, are discussed in the context of ligand and substrate effects. The review also addresses the scope, limitations, and synthetic applications of these reactions, with special emphasis on their utility in late-stage functionalization, heterocycle synthesis, and green chemistry. By summarizing the state-of-the-art in this evolving field, this review aims to provide a comprehensive reference for researchers interested in exploiting nitroarenes as aryl electrophiles in palladium-catalyzed cross-coupling chemistry.
