Expression and Characterization of Sucrose Phosphorylase from Thermanaerothrix daxensis and Production of 2-O-α-D-glucosyl glycerol
2-O--D-glucosyl glycerol (2-GG) is a valuable compound in cosmetics, health care, and food industries.Among various approaches, the enzymatic synthesis of 2-GG via the transglycosylation activity of sucrose phosphorylase (SPase) has been recognized as an efficient and promising strategy for its biosynthesis. However, only a limited number of SPases capable of catalyzing this reaction have been identified. Most of them exhibit limited stability at elevated temperatures, which highlights the need to identify novel enzymes with improved properties such as thermal stability. In this study, a novel SPase derived from Thermanaerothrix daxensis was employed to successfully synthesize 2-GG.Differential scanning fluorimetry (DSF) analysis revealed that the enzyme possesses high thermal stability with a melting temperature (Tm) of 79.4 o C, supporting its robustness under elevated temperatures. The catalytic properties of this biocatalyst were further evaluated under various pH conditions, and reaction parameters including temperature were optimized to enhance production efficiency.TdSPase exhibited optimal relative activity at pH 7.0 and 60 o C.These findings demonstrate that TdSPase is a promising biocatalyst for establishing an efficient biosynthetic platform with considerable potential for industrial application.
