Central Composite Design, Optimization, in-Vitro & Anti-diabetic Activity study of Repaglinide using lipid nanocarrier

The aim of this study was to create and assess a Repaglinide dispersion based on NLC. A drug's nanoformulation was prepared, and its characterisation parameters were calculated. Design expert software (central composite design) was used to optimize the current investigation. The concentration of cetyl alcohol, the concentration of oleic acid, and the influence of lecithin were the factors that were taken into consideration. The parameter that were taken into consideration were mean particle size and entrapment efficiency. By using the melt emulsification solidification at low temperature approach (with a small modification), repaglinide-encapsulated nanostructured lipid carriers were created. Characterizations included size distribution, drug entrapment efficiency, and NLC dispersion differential scanning calorimetry (DSC). The optimized batch was evaluated for in-vitro, pharmacodynamic studies of NLC encapsulated Repaglinide. The observed lipid matrix-NLC entrapment efficacy ranged from 76.28±0.72% to 86.64±0.64%, with significant particle sizes ranging from 254.4±12.2 nm to 464.43±14.7 nm. The formulation's long-term stability was also clarified by the Zeta potential assessment. SEM examines the particle's size distribution as determined by the Malvern equipment. The stability study's data also showed that the formulation remained stable after 180 days of storage. The in vivo antidiabetic evaluation shown that the blood sugar level may be lowered by Repaglinide-loaded NLC dispersion. Interestingly, in the case of the Repaglinide loaded NLC groups, and the average blood sugar values at all-time intervals were significantly less than that of the basal glucose value (p<0.05).
