Preparation of Magnetic Resonance Imaging Loaded Hemostatic Peptide Pentaerythritol Triacrylate Microspheres and Its Embolic Properties In Vivo
Transcatheter arterial chemoembolization (TACE) represents an interventional radiology technique that combines targeted chemotherapy with vascular occlusion. This procedure involves administering chemotherapy-impregnated embolic agents via microcatheters directly into tumor-feeding arteries, achieving dual therapeutic effects through sustained drug release and mechanical obstruction of the vascular supply to malignant lesions. In this study, pentaerythritol triacrylate (PETA) was used as the raw material, and PETA drug-loaded embolization microspheres with hemostatic, magnetic responsiveness, and magnetic resonance imaging abilities were prepared by microfluidic technology. Loaded peptide can accelerate blood coagulation at the embolization site and block blood flow in a short time. Successful arterial embolization was observed in rabbit models, with microsphere deployment achieving the desired hemodynamic obstruction. The multifunctional I3QGK@SPIONs/DOX@PETAMS composite demonstrates synergistic properties including enhanced magnetophoretic response, controlled drug release kinetics, rapid hemostatic action, contrast-enhanced visualization, and precise vascular occlusion, suggesting broad applicability in transarterial chemoembolization and related embolotherapy procedures.
