MULTIPLE SKLEROZ HASTALIĞININ DEĞERLENDİRİLMESİNDE ACKR2 GEN İFADESİNİN ÖNEMİ
Direct interactions between cells, such as those between the cell and the extracellular matrix, are extremely important for the development and function of multicellular organisms. Since peptide hormones, neuropeptides and growth factors do not have the ability to cross the plasma membrane of their target cells like cytokines, they bind to receptors on the cell surface such as ACKR2, which is a small cytokine. Therefore, since the ACKR2 receptor is a protein structure, gene expression is important in terms of functionality in the cell. A typical chemokine receptors (such as ACKR2, ACKR3); unlike chemokine receptors, are found in erythrocytes, lymphatic or vascular endothelial cells and especially ACKR2 and ACKR3 are detected in leukocytes. Therefore, for ACKR2 gene expression, leukocytes can be isolated by taking human whole blood and RT-PCR method can be used. Numerical findings in this gene expression will show differences in Multiple Sclerosis (MS) disease types. The aim of this study is to select the most appropriate gene for gene expression and to perform quantitative, molecular analyses correctly and to evaluate it according to the course of the disease. The time, condition and amount of gene expression will vary depending on the metabolic activities of the individual. The speed and condition of metabolic activity will also vary from person to person. This situation will help in making a personalized diagnosis when evaluated with the patients' living conditions, incontinence status, age, clinical observations, inflammatory and immunological data.
