[1]¿p#1 DEC205 as a Neutrophil Guidance Receptor: Unsupported Mechanistic Claims and Unverified Pathophysiological Relevance

The recent study by Jin et al. (Immunology.2025) presents the intriguing finding that a subset of bone marrow neutrophils expresses DEC205 and proposes a critical role for this receptor in guiding neutrophil migration to inflamed tissues. While this discovery nominates a novel player in neutrophil biology, our letter provides a forward-looking and substantive critique of the proposed mechanism. We identify significant gaps, including the absence of an identified ligand for DEC205 on endothelia, a lack of insight into the intracellular signaling cascades triggered by DEC205 engagement, and an undefined ontological status of DEC205+ neutrophils.Furthermore, we critically examine the pathophysiological relevance and highlight the substantial challenges in therapeutic translation, given the constitutive expression of DEC205 on dendritic cells and the potential for systemic immunomodulation. We conclude that while the findings open a new avenue of inquiry, the elevation of DEC205 to a validated neutrophil guidance receptor necessitates rigorous mechanistic and functional validation in diverse disease models.
