One-Step Synthesis of Interconnected Porous Molecularly Imprinted Polyampholyte Hydrogels via High Internal Phase Emulsions for Rapid and Selective Lysozyme Separation

The preparation of molecularly imprinted polymers by simple and universally applicable methods for the rapid and efficient purification of proteins has always been a significant challenge within the realm of biological separation. In this work, interconnected porous molecularly imprinted polyampholyte hydrogels (IPMIPs) were prepared by one-step free radical polymerization of monomers bearing opposite charges by using high internal phase emulsions as templates for lysozyme (Lyz) separation. By changing the concentration of surfactant Tween 20 and hydrophilic silica nanoparticles N20, the pore interconnectivity and pore size of the imprinted hydrogels could be finely adjusted, thereby affecting the swelling process. Owing to the well-defined and interconnected porous structure, IPMIPs were capable of achieving adsorption equilibrium within a remarkably short time frame of 45 min. The monolayer adsorption capacities of IPMIPs for Lyz reached an impressive 242.5 mg/g with an imprinting factor (IF) of 3.01 for Lyz. Through electrostatic complementation and hydrogen bond interactions, IPMIPs showed Lyz adsorption capacities significantly higher than those of other reference proteins, and the selectivity coefficient (α) to ovalbumin (OVA) was as high as 7.17. The real sample separation and regeneration experiments further demonstrated the excellent specific recognition ability and reliability of the IPMIPs. This strategy presents a highly promising alternative for the Lyz separation process and has the potential to be extended to other protein molecules.
