P05.19.A CLINICAL OUTCOME OF BIOMARKER-GUIDED THERAPIES IN ADULT NEURO-ONCOLOGY PATIENTS: AN UPDATE FROM THE TÜBINGEN MOLECULAR TUMOR BOARD COHORT

Abstract BACKGROUND Molecular profiling and target decision in the Molecular Tumor Board (MTB), within structured networks such as the Centers for Personalized Medicine (CPM), have broadened therapeutic options for neuro-oncology patients. Here, we provide an updated analysis of the Tübingen MTB cohort for adult patients with advanced tumors in the nervous system. MATERIAL AND METHODS The Tübingen MTB cohort is an ongoing observational study (NCT03503149) focusing on precision oncology in patients with advanced tumors. Upon enrollment, patients undergo comprehensive molecular profiling, and targeted therapy recommendations are recommended through interdisciplinary discussions in the MTB. RESULTS This report includes follow-up data on neuro-oncology patients up to the cutoff date of June 30, 2024. Of 1,088 patients enrolled, 854 were presented at the MTB (78.5%). Molecularly guided therapy was recommended for 773 of 854 patients (90.5%), with prioritization based on evidence levels. Treatment was initiated in 155 of 773 patients (20.1%). Among these, progression-free survival (PFS) data were available for 134 patients, of whom 47 (35.1%) achieved a PFS2/PFS1 ratio >1.3. CONCLUSION The CPM and MTB workflows have become an integral part of routine clinical practice and demonstrate clinical benefit in approximately one-third of treated patients.
