Antioxidant, Anti-inflammatory, and Wrinkle-Improving Effects of PDRN Extracted from Marine Plants

Purpose: This study aimed to determine whether polydeoxyribonucleotide (PDRN)-containing extracts from marine plants have antioxidant, anti-inflammatory, and/or wrinkle-improving effects. Methods: PDRN was extracted from a marine plant using a proprietary technology owned by our company. The antioxidant effect of marine plant PDRN was evaluated by quantifying 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, while its anti-inflammatory effects were assessed by quantifying NO inhibition in Raw264.7 cells. Finally, anti-wrinkle effects were evaluated through procollagen synthesis analysis in CCD-986sk cells, as well as by assessing epidermal layer thickness and collagen expression of the basement membrane through a 3D cultured skin model. In addition, the effects on the improvement of the four major facial wrinkles were verified in a human application test using a product formulated with marine plant PDRN.Results: The molecular weight of marine plant PDRN was 5–40 kDa, with a purity of 1.9, and cell viability remained above 80% at all concentrations. DPPH radical scavenging and NO inhibitory activities increased in a concentration-dependent manner. Procollagen production increased similarly, and the increases in epidermal layer thickness and basement membrane collagen expression in the 3D cultured skin model were similar to those of ascorbic acid. Wrinkle improvement was further confirmed using a human application test on the four major facial wrinkles. Conclusion: In this study we demonstrate that marine plant PDRN possesses strong antioxidant, anti-inflammatory, and wrinkle-improving effects, indicating that it may be suitable as a functional cosmetic ingredient.
