Stages of Seminiferous Tubules in Rats: Morphological Characterization by Light and Electron Microscopy and the Relevance of Staging

Objective: This study investigated testicular tissues from untreated control rats (rats that were not given any treatment or drugs) using light and electron microscopy. Seminiferous tubules were systematically classified into distinct stages according to the characteristic arrangement of spermatogenic cells. The objectives were to delineate these stages by light microscopy and to provide detailed morphological characterization at the ultrastructural level. Particular emphasis was placed on nuclear remodeling in spermatids, acrosome formation, and Sertoli cell associations across all 14 stages of the seminiferous epithelium. Electron microscopic analysis yielded further insights into spermatid differentiation, especially during acrosomal development and nuclear elongation. Collectively, these observations illustrate the dynamic progression of spermatogenesis and establish a comprehensive morphological framework that may serve as a reference for future investigations in reproductive biology and experimental testicular pathology. The primary objective of this study was to identify the 14 stages of rat seminiferous tubules by light microscopy and to characterize their morphological features in detail through electron microscopy. Material and Methods: Staging of seminiferous tubules was performed histochemically using periodic acid–Schiff (PAS) staining. Testicular tissue sections were further examined ultrastructurally with a transmission electron microscope to document stage-specific characteristics. Results: The 14 stages of the seminiferous epithelium were systematically defined according to their morphological features, documented by light and electron micrographs, and correlated with the spermatogenic cell series. A comprehensive table summarizes these stage-specific characteristics. Conclusion: This stage-based morphological atlas provides a valuable resource for elucidating the dynamics of spermatogenesis in the rat testis and may serve as a reference in reproductive biology and experimental pathology.
