Polymer–Silica Hybrid Hydrogels with Electrochemical Characteristics and Inhibit Diffuse Large B-cell Lymphoma Progression
Based on the research, a novel Zn(II)-based coordination polymer, [Zn(Hpzdc)(phen)] n (1), was synthesized using pyrazole-3,5-dicarboxylic acid (H₃pzdc) and 1,10-phenanthroline at ambient temperature, and subsequently encapsulated into a HA–APTMS@CP1 nanocarrier system to form HA–APTMS@CP1@1. Electrochemical analysis and pH-dependent release studies further confirmed the system’s responsive drug release behavior, with 86.8% cumulative release at pH 6.0 over 300 min, ensuring stability and targeting capability under tumor microenvironment conditions. Molecular docking simulations revealed strong interactions between compound 1 and key cell cycle regulatory proteins, suggesting potent biological activity. Diffuse large B-cell lymphoma (DLBCL) is a highly aggressive B-cell malignancy, and targeted interventions against cell cycle regulatory pathways may provide new strategies for treatment-resistant or relapsed cases. In vitro experiments using DLBCL cell line demonstrated that HA–APTMS@CP1@1 significantly inhibited cell viability after 48 h. Additionally, it reduced CDK6 and Cyclin D1 mRNA expression, indicating effective blockade of the G1/S cell cycle transition. Collectively, these findings highlight HA–APTMS@CP1@1 as a promising DLBCL-targeted delivery platform with excellent biological activity, controlled release properties, and strong therapeutic potential.
