STUDYING OF BIOCOMPATIBILITY INDICES OF CARBON MATERIALS FOR MEDICAL PURPOSES USING HUMAN MONONUCLEAR CELL CULTURE

Background. The development of modern medicine and the medical industry is associated with the development, growth of production and widespread adoption of carbon-based medical devices. This process requires more thorough selection and testing of the biocompatibility of carbon products, including using live cell cultures. Purpose. To conduct a comparative analysis of the biocompatibility of materials used for the manufacture of carbon implants and dressings using live human cell cultures. Materials and methods. The work uses mononuclear cells obtained from the blood of healthy donors. An extract was prepared from each carbon tissue sample according to GOST 32075-2013 and GOST 57467-2017. Cytotoxicity of extracts was tested using trypan blue. The proliferative reaction of cells was determined by the inclusion of H3 thymidine. The effect of extracts on the production of interleukins 2, 4, 6 and tumor necrosis factor alpha was studied in the cell culture. Results. It was found that the reaction to different carbon fabrics was also different. Cytotoxicity, stimulating cell proliferation and cytokine production, was practically absent in the Carpema tissue, while Carbotextim-M carbon felt had some cytotoxicity significantly different from the control, as well as a pronounced tendency to stimulate cytokine production and cell proliferative activity. Conclusions. The revealed mitogenic and cytokine-stimulating properties of carbon tissues indicate the expediency of improving methods for testing the biocompatibility of medical materials, including using human cell cultures.
