Cyclodextrin-based supramolecular polymers based on host–guest interactions for cancer therapy

Cyclodextrin-based supramolecular polymers (SPCDs) have emerged as promising platforms for cancer therapy via the utilization of cyclodextrin (CD)-mediated host-guest interactions and supramolecular chemistry. The unique molecular structure of CDs enables efficient encapsulation of therapeutic drugs in hydrophobic cavities. These host-guest interactions facilitate the construction of stimulus-responsive delivery platforms, allowing therapeutic cargoes to be released in the presence of endogenous (pH or enzymes) or external stimuli (light or temperature). In this review, we summarize the classification of SPCDs and highlight their representative applications in cancer treatment. In addition, we also discuss the current challenges and prospects for promoting the development and clinical translation of SPCDs. • Cyclodextrin-based supramolecular polymers (SPCDs) with various structures are introduced. • Recent developments in the use of SPCDs in a variety of tumor treatments, including chemotherapy, phototherapy, chemodynamic therapy, immunotherapy, gene therapy, and protein delivery, are summarized. • The promising prospects and potential challenges of the use of SPCDs in cancer therapy are outlined.
