Effects Of Combined Ethanolic Extracts of Citrullus Colocynthis and Plumbago Zeylanica (L) On Mcf-7 Human Breast Cancer Cell Lines
Background: Breast cancer is most common disease in both developed and developing countries. According to the World Health Organization, breast cancer is the most prevalent cancer among women worldwide and the second most common cause of cancer death inwomen. Breast cancer globally with over 1 million new cases diagnosed annually, resulting in over 400,000 annual deaths and about 4.4 million women living with breast cancer. Aim: Therefore, the aim of the study was investigate the effect of combined ethanolic extract of citrullus colocynthis and plumbago zeylanica linn on MCF-7 human breast cancer cell line. Methodology: The phytochemical screening performed combined ethanolic extract of citrullus colocynthis and plumbago zeylanica linn that presence of alkaloids, terpenoids, flavonoids, tannins and coumarins. GC-MS analysis was performed using thermo GCTrace Ultra Version 5.0 to determine the possible chemical compound .Molecular docking was performed for target protein 3ERT retrieved from the RCSB PDB. MTT assay performed to determine the anti-proliferative activity of combined ethanolic extract with different concentration such as 6.25, 12.5,25,50,100 µg/ml on MCF-7 human cancer cell line. Result: GC-MS analysis revealed 16 chemical component identified. In that 9-octadecenamide 50.43%, 1,2 benzenedicarboxylic acid, dioctyl ester 11.25% . Molecular docking study proved that the polyphenolic compound good binding affinity with glide score range of -9.85 kcal/mol to -5.98 kcal/mol compared to tamoxifen with glide score range of -10.28 kcal/mol.Cytotoxicity against MCF-7 cell line determined at the concentration of 25 µg/ml.Conclusion: According to the results, we conclude that combined ethanolic extract of citrullus colocynthis and plumbago zeylanica linn act as potential chemotherapeutic agent for breast cancer.
