NaGdF4@PDA-ALD Nanoparticles Enhance Mesenchymal Stem Cell Therapy for Ischemic Stroke: MRI Tracking and Protection Mechanisms

Motivation: Ischemic stroke limits the survival of transplanted mesenchymal stem cells (MSCs) due to oxidative stress and calcium overload in the ischemic microenvironment. Goal(s): The study aims to improve MSC survival and therapeutic efficacy in stroke therapy by shielding MSCs using functional nanoparticles. Approach: NaGdF4@PDA-ALD nanoparticles (NPANs) were synthesized to protect MSCs from reactive oxygen species (ROS) and calcium overload, while enabling in vivo tracking via MRI. Results: NPAN-treated MSCs showed enhanced survival, migration to the stroke area, improved blood flow, and angiogenesis, confirming the potential of NPANs to enhance MSC-based stroke therapies. Impact: This study provides a novel strategy to enhance MSC-based therapies for ischemic stroke, potentially improving patient outcomes. The nanoparticles protect MSCs might open new avenues for developing more effective cell therapies and in vivo tracking methods for regenerative medicine.
