Macromolecular Proton Fraction Mapping of Subcortical Nuclei in Patients with Parkinson's Disease

Motivation: Imaging biomarkers are needed to detect neurodegeneration in the prodromal stage and predict disease progression in Parkinson's disease. Goal(s): To assess the utility of macromolecular proton fraction (MPF) mapping and quantitative susceptibility mapping (QSM) of subcortical nuclei for diagnosing Parkinson's disease. Approach: We performed MPF mapping and QSM in patients with Parkinson's disease and healthy controls. Results: In multiple subcortical nuclei, patients with Parkinson's disease showed significantly decreased MPF values and significantly increased magnetic susceptibility values compared with that in healthy controls. Impact: Decreased MPF may reflect neurodegeneration, while increased magnetic susceptibility suggests iron accumulation. Combining MPF and QSM measurements in subcortical nuclei could aid in diagnosing Parkinson's disease.
