Near-infrared fluorescence imaging with a MET-targeting probe for biopsy site selection in patients with oral potentially malignant disorders

Motivation: Early detection of malignant transformation in oral potentially malignant disorders (OPMDs) is challenging. Goal(s): This study evaluates a MET-targeted near-infrared fluorescence imaging (NIRFI) probe, cMBP-ICG, to improve biopsy site accuracy in OPMDs. Approach: Using preclinical models and a clinical trial with 50 patients, NIRFI-assisted biopsy was compared to conventional methods. Results: Results showed NIRFI achieved 100% sensitivity and 86% specificity in detecting high-grade dysplasia and early malignancies, outperforming traditional biopsy accuracy (91% vs. 72%). These findings indicate that NIRFI has potential to enhance diagnostic precision and support earlier intervention for patients at risk of oral cancer. Impact: This study highlights NIRFI-assisted biopsy as a promising tool for enhancing early detection in oral cancer risk. By improving diagnostic precision in OPMDs, it opens avenues for research into broader applications of NIRFI in other head and neck cancers.
