Remodeling Of Glycerophospholipid Metabolism And Exercise Anti-inflammatory Effect Of Liver Tumor

PURPOSE: The relationship between inflammation and abnormal lipid metabolism in the anti-tumor effect of exercise is still unclear.,this study aims to investigate whether the input of inflammatory inducer lipopolysaccharide after exercise impact the anti-tumor effect of exercise. METHODS: 3-week-old C57 male mice were randomly divided into control group (NC group), injected with normal saline and did not exercise; HCC model group (DEN group) was treated with intraperitoneal injection of Diethylntrosamine (DEN) to induce HCC and did not exercise. DET group, DEN was injected to induce HCC, and aerobic exercise treatment was performed; Lipopolysaccharide (DETL) group was induced HCC model and then followed by injection of lipopolysaccharide after aerobic exercise. Immune function, cell apoptosis was detected by immunohistochemistry and immunofluorescence, and inflammation and proliferation-related indexes were detected by enzyme-linked immunoassay, RT-qPCR and WB. The liver lipid metabolites were detected by UPLC-QEMS technique. RESULTS: Compared with DEN group, DET group significantly decreased the incidence of liver tumor, inflammatory cell infiltration. The mRNA expressions of SOD1and PGPX4 in DET group were up-regulated (p < 0.001, p < 0.001). Aerobic exercise decreased serum IL-1β and TNF-α (p < 0.01, p < 0.01) and MCP1and CSF1 mRNA in liver (p < 0.01, p < 0.05). Compared with DET group, serum IL-1β and TNF-α were significantly increased in DETL group after exogenous lipopolysaccharide input after exercise (p < 0.01, p < 0.05). Western Blotting showed that NLRP3protein was almost unexpressed in DETL group. Compared with DET group, the number of Tunel cells in DETL group was significantly decreased (p < 0.01), and serum AFP-L1, AFP-L3 and VEGF were increased (p < 0.001,p < 0.01,p < 0.001). Lipidomics analysis showed that glycerophospholipid metabolism is a differentiated metabolic pathway under post-exercise inflammatory stress. CONCLUSION: The inflammatory pressure after exercise leads to abnormal glycerophospholipid metabolism, which will impair the anti-tumor effect of aerobic exercise on liver. Glycerophospholipid metabolism pathway is the main metabolic pathway of lipid remodeling in liver during exercise under lipopolysaccharide inflammatory pressure.
