Sum-frequency generation imaging of tissue proteins
We apply sum-frequency generation (SFG) microscopy to study the molecular structure of the extracellular matrix in tissues. By polarization-resolving individual elements of the nonlinear susceptibility tensor, we visualize structural differences in tissue collagen based on contrast derived from hydroxyl stretches and amide-specific vibrational modes.
