GSH-responsive magnetic imaging for <i>in situ</i> monitoring of tumor chemodynamic therapy

Chemodynamic therapy (CDT) is a promising treatment modality that utilizes highly toxic hydroxyl radicals (˙OH) to kill tumor cells <i>in situ</i>. However, real-time monitoring of drug distribution and therapeutic response remains a challenge, hindering the optimization of personalized treatment regimens. Here, we developed a glutathione (GSH)-responsive nanoreactor (FA-Mn NR), with integrated magnetic resonance imaging (MRI) functionality for self-enhanced CDT. The nanoreactor undergoes GSH-triggered Mn2+ release, which simultaneously functions as an MRI contrast agent and catalyzes hydrogen peroxide (H2O2) to produce additional ˙OH, thereby achieving synergistic CDT amplification. The study demonstrates a theranostic platform combining GSH-responsive imaging guidance with enhanced therapeutic efficacy, presenting a novel approach for cancer therapy.
