Development of an Efficient Bioconversion Strategy for Transforming Naringin from Pomelo Thinning Waste into Dihydrokaempferol

Pomelo (Citrus maxima) thinning, generated during early fruit development in southern China, is typically discarded due to the low economic value of conventional naringin extraction. However, these byproducts contain higher concentrations of 2S-naringin compared to mature pomelo. This study presents an efficient bioconversion strategy for transforming 2S-naringin from pomelo thinning waste into dihydrokaempferol, a pharmacologically active flavonoid. Optimized ultrasonication-assisted extraction yielded 17.2 g naringin with the 2S/(2S + 2R) ratio of 88.2% from 1000 g of pomelo thinning. Subsequently, AmRha was coupled with BGL-NL to prepare naringenin from naringin. Under optimal conditions, 5 mM naringin was converted into 4.85 mM naringenin after a 2 h reaction with a corresponding molar conversion of 97%. Finally, a total of 7.8 g naringenin with the 2S/(2S + 2R) ratio of 86.8% was prepared from 1000 g of pomelo thinning, demonstrating an efficient preparation method for obtaining naringenin with high 2S/(2S + 2R) ratio from pomelo thinning. Citrus sinensis flavanone 3-hydroxylase (CsF3H) was screened to construct the recombinant strain to produce dihydrokaempferol from 2S-naringenin. By optimizing conversion conditions and adopting the fed-batch fermentation strategy, maximal dihydrokaempferol production reached 1468 mg/L at 24 h after induction, with a corresponding molar conversion of 99% for (2S)-naringenin. Ultimately, 7.01 g dihydrokaempferol was prepared from 1000 g of pomelo thinning. This work provides an efficient approach for improving the added value of pomelo thinning with an environmentally safe process.
