Integrated Analysis of Super-Enhancers and Gene Expression Reveals Regulatory Networks in Ovarian Cancer
Ovarian cancer remains one of the most lethal gynecologic malignancies, partly due to the lack of early detection and effective therapeutic targets. Super-enhancers (SEs) are large regulatory clusters that drive high transcription of oncogenic genes. In this study, we constructed a super-enhancer landscape using H3K27ac ChIP-seq data from 17 ovarian-related cell lines. Differential SE analysis revealed widespread enhancer reprogramming, with cancer-specific SEs enriched in pathways such as Wnt signaling and chemokine signaling. By integrating SE activity with gene expression, we identified two master regulators, KLF5 and ZBED4. Further regulatory network analysis prioritized ZNF816 as a candidate druggable node that interacts with multiple FDA-approved drugs. These findings suggest that SE-associated regulators and their transcriptional targets may offer opportunities for therapeutic exploration in ovarian cancer.
