Time Modulated Illumination for Single Molecule Localization Microscopy
In Single Molecule Localization Microscopy (SMLM), the positions of the fluorophores are obtained from a fitted Point Spread Function. This spatially based localization precision will then strongly depend on the PSF shape, which can be degraded by defocusing and aberrations and affect both lateral and axial localization precision, but also the capability to image in depth in complex samples.
