H-MnO2 nanoparticles loaded with photosensitizer IR825 target the tumor microenvironment to alleviate hypoxia and synergy therapy

Tumor microenvironment (TME) is characterized by low pH, high hydrogen peroxide (H 2 O 2 ) concentration and severe hypoxia, which lead to tumor progression, metastasis and drug resistance. The development of TME-responsive smart nanosystems has shown great potential to improve the efficiency of cancer therapy. Metal Mn has the characteristics of catalase and can play the therapeutic effect of CDT. The synergistic treatment strategy of chemodynamic therapy combined with photothermal therapy and photodynamic therapy can play a better anti-tumor effect. The acidic conditions in the tumor microenvironment can promote the production of oxygen from H 2 O 2 , improve the anaerobic state of tumors, and provide oxygen materials for photodynamic therapy and chemodynamic therapy to produce more ROS. Therefore, we constructed a multifunctional nanocarrier system with photothermal therapy, photodynamic therapy and chemokinetic therapy for the treatment of prostate tumors. Scheme: Mechanistic diagram of the anti-tumor effect of high levels of cytotoxic ROS produced by the combined PTT/PDT/CDT regimen mediated by H-MnO 2 @IR825 under laser irradiation.
