Synergistic Effect of Irisin and Rapamycin in Diabetes Complicated Cerebral Ischemia

Background: Cerebral ischemia, a predominant cause of stroke, results from a reduction in cerebral blood flow, leading to neuronal injury and brain dysfunction. Diabetes mellitus (DM), a metabolic disorder characterized by chronic hyperglycaemia, significantly elevates the risk of ischemic stroke. Remote Ischemic Preconditioning is a promising strategy to recover the loss of the reperfusion injury Methods: A total 60 Wistar rat were employed for the current research and 10 group(n=6) on the impact of diabetes on cerebral ischemia, streptozotocin (50mg/kg) was used to induce diabetes. Bilateral common carotid artery occlusion model was adopted for the cerebral ischemia with focusing on the underlying mechanisms, clinical implications, and potential therapeutic approaches. Elevated Plus maze, Moris water maze were used for learning and spatial memory. Several parameters such as TBARS, superoxide anions, serum nitrite/ nitrate were measured for the oxidative stress histopathology were examined for the cerebral infraction by using TTC staining dye. statistically analysis was completed by using one-way ANOVA (sigma stat 12.5) Results: no significant variation were found in the sham group, vehicle perse1 and 2 group body weight and FBG were significantly attenuated in the diabetes with BCCAO group 210±6.9 the oxidative parameters were elevated in the disease control mellitus contributes to cerebral ischemia through several mechanisms, including endothelial dysfunction, increased oxidative stress, and chronic inflammation. STZ-administered rats showed reduction in body weight, learning, memory, reversal learning, executive functioning, increase in serum glucose, brains’ oxidative stress, inflammation, and histopathological changes. Administration of irisin and rapamycin alone as well as in combination, significantly and dose dependently attenuated the STZ-diabetes-induced impairments in the behavioural, endothelial, and biochemical parameters. Conclusion: STZ administration caused diabetes with bilateral common carotid artery occlusion (BCCAO) complicate the cerebral ischemia whereas the protective effect of RIPC was significantly restore and attenuated by the administration of irisin and Rapamycin in combination Therefore, these agents may be studied further for the assessment of their full potential in diabetes induced cerebral ischemia
