pH-Responsive Proteomimetic Nanodelivery System for the Diagnosis and Treatment of Anaplastic Thyroid Carcinoma

Anaplastic thyroid carcinoma (ATC) currently lacks satisfactory diagnostic strategies and treatment plans. Traditional anti-cancer drugs, due to their lack of targeting, are prone to causing systemic toxicity, which greatly limits their clinical application. Therefore, we developed a new and effective targeted drug delivery system, MnO 2 @HSA-DOX, which can enhance the targeting of drugs in the tumor area and reduce the impact of drugs on normal tissue cells. The average particle size of this nanoparticle is 86.48[Formula: see text]nm, and it can smoothly reach tumor cells. Due to its characteristic of responding to the acidic tumor microenvironment (TME), it can rapidly degrade and release in the TME, releasing Mn[Formula: see text] and DOX, thereby achieving integrated diagnosis and treatment. In vitro experiments have demonstrated that albumin nanoparticles as drug delivery carriers have good biological safety, and MnO 2 @HSA-DOX has a better therapeutic effect than free Doxorubicin (DOX). This result indicates that this nanoprobe can precisely deliver drugs to thyroid cancer cells for precise treatment. In vivo experiments have shown that this probe has low cytotoxicity and can clearly display tumor tissue in the magnetic resonance imaging (MRI) of the 8305C BALB/c nude mouse model. This innovative nanomaterial provides a comprehensive diagnostic platform that combines therapeutic and imaging capabilities. Therefore, it is considered a way to promote cancer diagnosis and treatment.
