Study of dermal toxicity of SPT hemostatic dressing during long-term use

The article presents the results of the study of dermal toxicity of the hemostatic dressing SPT (gradients: alginate chitosan wood (AHD), glycerin, and PEG 400) during long-term use. Healthy, young females with intact skin, weighing 230-250 g, were used for the experiment. The study of dermal toxicity of the studied agent during long-term use was carried out following OECD No. 410 (Repeated Dose Dermal Toxicity: 21/28-day Study). For this purpose, 4 groups of animals were formed according to the principle of analogues, five rats in each. Water was applied to the previously prepared area of skin of the animals of the control group, 0.1 ml/kg of the animal's body weight (b.w.) for the I experimental group, 0.25 ml/kg of the II experimental group, and 0.5 ml/kg of the III experimental group. 28-day cutaneous application of the drug to females of the experimental group I led to an increase in the weight coefficient of the liver, spleen, heart and kidney mass, an increase in hemoglobin concentration, the number of erythrocytes, the level of hematocrit, platelets, lymphocytes, total protein, albumin, creatinine level, as well as the activity of AST, ALT, LDH, cholesterol, calcium and phosphorus levels. At the same time, a decrease in the number of leukocytes, the average hemoglobin content in erythrocytes (MCH), the average erythrocyte volume (MCV), as well as a decline in the number of monocytes, granulocytes, the level of urea, alkaline phosphatase (ALP), triglycerides (TAG), CL and bilirubin was observed. 28-day cutaneous application of the drug to females of the II experimental group caused an increase in the weight coefficient of the liver, spleen, heart and kidneys, body weight, as well as an increase in hemoglobin concentration, number of erythrocytes, leukocytes, hematocrit level, platelets, mean hemoglobin content in erythrocytes (MCH), mean erythrocyte volume (MCV), lymphocytes, albumin, creatinine level, activity of AST, ALT, LF, LDH, cholesterol, TAG and calcium level. At the same time, the mean hemoglobin concentration in erythrocytes (MCHS) decreased, and a decrease in the number of monocytes, granulocytes, total protein, urea, CL, and bilirubin was observed. 28-day cutaneous application of the drug to females of the III experimental group led to an increase in the weight coefficient of the liver, spleen, heart and kidney mass, body weight, number of erythrocytes, hematocrit level, platelets, lymphocytes, albumin, creatinine level, activity of AST, ALT, LF, LDH, cholesterol, TAG, calcium, chlorine and bilirubin levels. At the same time, a decrease in hemoglobin concentration, number of leukocytes, mean hemoglobin content in erythrocytes (MCH), mean hemoglobin concentration in erythrocytes (MCHC), mean erythrocyte volume (MCV), number of monocytes, granulocytes, total protein, and urea level was observed.

Abstract

The article presents the results of the study of dermal toxicity of the hemostatic dressing SPT (gradients: alginate chitosan wood (AHD), glycerin, and PEG 400) during long-term use. Healthy, young females with intact skin, weighing 230-250 g, were used for the experiment. For this purpose, 4 groups of animals were formed according to the principle of analogues, five rats in each.
