In-flow digital holographic tomography for 3D phytoplankton visualization and characterization
The study and characterization of phytoplankton is of primary interest as they play a fundamental ecological role and constitute bioindicators for climate changes. Indeed, human-induced activities alter their morphology and impact their colonies. The state-of-the-art microscopy techniques allow the high-throughput visualization and enumeration of 2D images of phytoplankton and the estimation of basic parameters; however, their 3D imaging and characterization is a challenging task due to their extremely variegated morphologies that has not been fully investigated. Here, we propose for the first time in-flow digital holographic tomography as a quantitative, label free technique to recover the 3D refractive index distribution of phytoplankton, addressing the challenges related to their non-homogeneous rotations in microfluidic environment.
