Development and Validation of a UV-Visible Spectrophotometric Method for Simultaneous Estimation of Curcumin and Clindamycin

This study aimed to develop and validate a simple, accurate, and cost-effective UV-visible spectrophotometric method for the simultaneous estimation of curcumin and clindamycin in bulk drug form. Pre-formulation studies including organoleptic evaluation, solubility analysis, pH, and melting point determination were performed. UV absorbance maxima (λmax) were determined to be 420 nm for curcumin and 202 nm for clindamycin. A simultaneous equation method and isoabsorptive point method at 262 nm were employed for estimation. The method was validated as per ICH Q2(R1) guidelines for parameters such as linearity, precision, ruggedness, robustness, and drug-excipient compatibility via FTIR. Preformulation studies confirmed organoleptic properties, solubility, and compatibility. The selected solvent system was water:methanol. Curcumin and clindamycin showed λmax at 420 nm and 202 nm, respectively. The method followed Beer’s law within specified concentration ranges and showed excellent linearity (R² ≈ 0.998). Validation as per ICH guidelines confirmed precision, ruggedness, and robustness. The validated UV-visible spectrophotometric method is accurate, precise, and suitable for the routine simultaneous estimation of curcumin and clindamycin in pharmaceutical formulations.
