Phylogeny of Three Species Belong to Genus Potyvirus Infecting Solanum tuberosum Based on Biological and Molecular Variability

Potato virus Y exhibits diverse strains and complexities that circulate within potato fields and decrease potato tuber yields. In the present study, conducted from 2021 to 2023 across various seasons and regions, Potato Virus Y (PVY) isolates were examined for their diverse symptom expressions on potato plants. The infected potato plants were categorized into three distinct phenotype expressions, venial necrosis (PVY-VN), yellow mosaic (PVY-YM) mottling pattern (PVY-MP), they showed different biological factors such as disease density index, pathogenicity, virus concentration by DAS-ELISA, and molecular variability through HC-PRO silencing gene via qRT-PCR. The phylogeny of HC-PRO silencing gene nucleotide sequences composition (207 bp) of three PVY isolates showed differed nucleotide sequences similarly and genetic distance between them and other PVY strains depended on Genbank database and recorded on the Genbank under accession numbers OR804503, PQ015611, and PQ015612 of PVY-VN, PVY-YM, and PVY-MP respectively. Depending on the phylogeny, the three isolates belonged to the NTN strain ranged 99% to 100% sensitivity with AJ585342_United-Kingdom-strain_NTN sequence and KY092173_Poland_strain_sequence.
