Complete Genome Sequence of an optrA-Positive Linezolid-Susceptible Staphylococcus simulans Strain PFJB-50N Isolated from a Pig Farm
The occurrence of linezolid resistance in non-aureus staphylococci (NAS) isolated from pig farms has posed a significant threat to public health [1].Acquisition of optrA gene, which usually confers transferable resistance to oxazolidinones (linezolid and tedizolid) and phenicols (chloramphenicol and florfenicol), have been observed in isolates of enterococci and staphylococci derived from humans and food-producing animals [2].However, in this study, we present the complete genome sequence of an optrA-positive but linezolid-susceptible S. simulans strain, PFJB-50N, isolated from a pig farm in Korea. The S. simulans PFJB-50N strain was isolated from a swab sample collected from a feeder box in a pig farm located in Jeolla-do, Korea. For whole-genome sequencing analysis, genomic DNA sample of the PFJB-50N strain was extracted using the Wizard Genomic DNA Kit (Promega, USA).The construction of sequencing Linezolid, a synthetic antibiotic in the oxazolidinone class, has been approved for human clinical use since 2000.However, recent emergence of oxazolidinone-resistant Staphylococcus spp.carrying optrA gene, encoding an ATP-binding cassette-F protein, in pig farms has raised significant public health concerns in Korea.Although the presence of optrA gene has been associated with linezolid-resistant phenotype in Staphylococcus spp.and Enterococcus spp., an optrA-positive linezolid-susceptible Staphylococcus simulans strain PFJB-50N was isolated from a pig farm in Korea.Sequence analysis of the optrA gene in PFJB-50N revealed five nucleotide point mutations associated with five amino acid substitutions.
