Inhibition of Skin Inflammation and Serum Cytokine Production by Herbal Complex Extracts

This study studied the effect of S-E(Cinnamomum japonicum, Japanese Ampelopsis, Atractylodes macrocephala Koidz, Poria Cocos, Bletilla striata (Thunberg) Reichenbach fil, Agastrche rugosa O.KTZE, Cinnamomum japonicum) on suppressing skin inflammation in DNCB-induced NC/Nga mice and obtained the following conclusions. To confirm the effect of S-E on suppressing skin inflammation, contact dermatitis was caused in NC/Nga mice using DNCB, and the skin clinical index was checked, showing a significant decrease of 45.46% compared to the control group (DNCB-treated group). In addition, as a result of confirming the effect of S-E on skin tissue with H&E staining and T&B staining, inhibition of epidermis and dermis edema and leukocyte infiltration in the skin were confirmed, and the number of mast cells around the dermis was decreased, indicating that damage to the skin barrier caused by stimulation of mast cells was reduced. IgE, which plays an important role in allergic reactions, combines with mast cells and basophils to induce allergic reactions, affecting skin inflammation. Accordingly, in order to investigate the effect on the expression of IgE, blood was collected from the eyes of NC/Nga mice at 12, 14, and 16 weeks, the serum was separated, and the amount of IgE production was measured. As a result, the S-E showed a significant reduction of 3.4%, 38.3%, and 48.84% compared to the control group. As a result of measuring the effect of S-E application on the amount of cytokine production in the serum in NC/Nga mice with skin inflammation, IL-6 production was significantly reduced by 38.38% compared to the control group, and TNF-α production was also significantly reduced by 37.5%, suggesting that it is effective in suppressing the development of skin inflammation. Therefore, the results of this study indicate that S-E is effective in suppressing dermatitis in NC/Nga mice induced by DNCB, and it is considered worthy of active use and various studies in future research on materials related to dermatitis.
