The CALCIPHYX trial and its impact on calciphylaxis treatment: what's next?
Calciphylaxis, also known as calcific uremic arteriolopathy, is a rare vasculopathy characterized by calcium deposi on in the arterioles of the deep dermis and subcutaneous adipose ssue that results in severely painful, ischemic skin lesions [1].The pathogenesis of calciphylaxis is not yet fully elucidated.Medial calcifica on of arterioles in the deep dermis and subcutaneous adipose ssue is proposed to play a crucial role, leading to ischemia, endothelial injury, ssue infarc on, and skin necrosis (Figure 1) [1].Although calciphylaxis is associated with several complica ons ( ssue necrosis, infec ons, sepsis, etc.) and extremely high mortality rates (40-70% at 1 year) [2,3], there are no approved therapies [1].
