Development of Reflective Biosensing Probes for In Situ Biological Monitoring

In medical diagnosis and life science research, real-time in situ detection of biological samples is important for early disease screening and dynamic physiological monitoring. The limitations of traditional bioassay technologies, such as complex pre-processing steps, time-consuming detection, and inability to achieve real-time in situ monitoring, significantly limit their potential applications in in vivo tissue analysis and immediate clinical diagnosis. In this paper, we present a low-cost and easy-to-prepare reflective biosensing probe for monitoring biochemical processes recognized by protein molecules. The results show that the refractive index sensitivity of our designed probe reaches 2373.4 nm/RIU. The fiber-optic probe was functionalized using a covalent bond coupling method, and target capture was achieved by combining the antigen-antibody specific recognition mechanism. The signal response regeneration rate still reached 94.58% after three regeneration cycles. The wavelength shift of the sensing probe has a good linear relationship with the concentration of the protein molecule, and the design of this reflective sensing probe has an important application value in the practical application scenario of medical detection.
