Motility-based scrutiny of probiotic bacteria in vitro digestive environment achieved by an all-optical platform
We propose an all-optical platform, which contains two different optical measurement methods: digital holography (DH) and bio-speckle decorrelation analysis (BSDA), for profiling bacteria strains that are candidates to serve as probiotics. BSDA allows global evaluation under large Field of View (FOV) at the cost of lateral resolution; DH under high magnification can achieve 3D tracking of individual bacteria but the FoV is extremely limited. The complementary properties of these two approaches allowed us to perform a comprehensive analysis of bacterial motility. Different bacteria are encapsulated in alginate microcapsules separately. They will be put in solutions with different pH to simulate in vitro digestive environments. As a preliminary result, quantitative analysis of the motility of different strains of lactic acid bacteria are presented.
