Application of Electrospun Antibacterial Nanofibrous Membranes in Skin Wound Repair

Skin injuries and defects in the head and face are clinically tricky problems. These infected skin defects are more difficult to repair due to the specificity of their location. Therefore, in this study, a nanofibrous membrane with antimicrobial properties was developed to repair full-layer skin defects. Briefly, ZnO was organically combined with eggshell powder through the adhesion of dopamine to prepare eggshell-polydopamine-ZnO (ES-PDA-ZnO) composites. Then, the antimicrobial nanofibrous membranes were spun into antimicrobial nanofibrous membranes by electrostatic spinning, which were used to mimic the epidermal tissues of the skin. To enhance the antioxidant function of the material and promote wound healing, some fish scale gelatin solution was doped to electrostatic spinning. The compositions of the composites were subsequently identified and analyzed by tests such as X-ray diffraction analysis (XRD), Fourier transform infrared spectroscopy (FT-IR), and X-ray photoelectron spectroscopy (XPS). The antimicrobial properties of the materials were verified by in vitro antimicrobial tests, and the hydrophilicity of the nanofibrous membranes was also investigated by water contact angle measurements. Meanwhile, the nanofibrous membranes were verified to have good biosafety by an in vitro cytotoxicity test. The cell scratch test indicated that the nanofibrous membrane promotes cell migration, which is a key factor in wound healing. These in vitro findings suggest the potential for further exploration in skin regeneration, pending validation through in vivo studies.
