Solubility Enhancement of Nateglinide by Using Gelucire

The aim of the present work is to investigate the possibility of obtaining immediate release tablet of nateglinide is used to treat type-II diabetes as it belongs to BCS-II i.e, have low solubility and high permeability. The immediate release of nateglinide with improved dissolution using solid dispersion and crystalline solid formulation, amorphous formulation and lipid formulation techniques the solubility and dissolution rate of nateglinide can be enhanced by solid dispersion of nateglinide with gelucire 44/14 reduction of particle aggregation of the drug, formulation of amorphous drug increased wettability and dispersibility The neteglinide immediate release table F9 showed 78.48% drug wihin 40min and 98.36% drug release within 60 min.The results that he formulation satisfied he objective of dissolution and hardness, bulk density, tapped density within the limits of nateglinide drug with different gelucire grades like 43/01, 50/13, 39/01, 44/14, 33/01,48/16 a strong enhancement of drug dissolution rate was obtained from the nateglinide: gelucire 44/14 by using co-precipitation method and hydrophobic excipient which allows it to self-emulsifying in water, forming a fine dispersion when coming into contact with a aqueous media making it useful for enhancing the solubility of poorly water soluble drug.
