TECHNOLOGICAL RESEARCH IN THE DEVELOPMENT OF A RECTAL DOSAGE FORM OF A COMBINED COMPOSITION
The pharmaceutical market has a limited range of dosage forms with modified pharmacokinetic properties. The aim of the work is to develop a promising drug of combined pharmacological action based on metronidazole and oil extract of Tambukan mud in the form of two-layer rectal suppositories. The object of the study was suppositories based on metronidazole and oil extract of Tambukan mud. The research methods were based on pharmacopoeial techniques, as well as tests using the Kruvchinsky method. The bioavailability results demonstrate that the proposed bases used in the development of the optimal composition of suppositories with metronidazole do not interfere with its release. From a biopharmaceutical point of view, the optimum release of active components from the outer layer is provided by the combined polyethylene oxide base PEO – 400 : PEO - 1500 in the ratio (95:5), and from the inner layer, the optimal profile for metronidazole is observed when using Suppocire CS2X solid fat. Two–layer suppositories were obtained by pouring: an outer layer based on polyethylene oxides with an extract from Tambukan mud, and a core layer based on Suppocire CS2X with metronidazole introduced by suspension type. The authenticity of the active pharmaceutical substances was verified by the spectrophotometric method at a wavelength of 312 nm. Conclusions. The proposed two-layer suppositories based on metronidazole and oil extract from Tambukan mud will enable the development of controlled drug delivery with a modified release profile, through the use of layers of different compositions, which will combine layers with gradual and immediate release. These suppositories will improve the effectiveness of infection treatment, providing an anti-inflammatory effect and speeding up the patient's recovery process.
