Abstract 2914 Role of RNase L in the Spike Protein of SARS-CoV-2 (S-protein)-Induced Activation of Macrophages

High-throughput single-cell RNA sequencing (scRNA-seq) has revolutionized our understanding of RNA molecules and has enabled quantitative analysis of gene expression. In the context of T cells, scRNA-seq can be used to analyze the T cell receptor (TCR) repertoire, which provides insights to T-cell antigen specificity.Despite its potential, the widespread application of scRNA-seq is often constrained by its high cost and the susceptibility of RNA degradation in the sample during processing. To address these limitations, we focused on optimizing the encapsulation efficiency and RNA preservation in a non-microfluidic scRNA-seq method called particle-templated instant partition sequencing (PIP-seq), using T cells in the experimental system.Variables such as oil volume, cell volume, and bead-to-cell ratios were tested to improve the T cell and bead encapture rate.Additionally, recovery conditions were tested to minimize TCR degradation and ensure higher quality input for downstream sequencing. This work demonstrates the foundation for the feasibility of a more cost-effective scRNAseq technique for the analysis of T cells.
