Abstract 2108 Cobalt Bead-Based Selection for a Therapeutic Aptamer Against Mutated Superoxide Dismutase 1 Alleviates Multiple Sclerosis Symptoms via M1 Muscarinic Inhibition

Mitochondrial genome plays a central role in assuring production of ATP and meeting energetic requirements of the cell.Because of this critical role, numerous human neuromuscular or metabolic disorders are associated with instability of mitochondrial DNA (mtDNA).We have recently observed that overexpression of mitochondrial chaperone factors promoted quantitative and qualitative condition of mtDNA.This indicates that mobilization of mitochondrial chaperones could be a potential avenue for therapeutic interventions in the mtDNA associated disorders. This, however, depends on finding applicable means of elevating cellular levels of mitochondrial chaperones.Notably, recent studies indicated that application of ganoderma microsporum immunomodulatory proteins (GMI) can activate mitochondrial chaperones and, in turn, improve mitochondrial homeostasis. Here we sought to assess whether supplementation with GMI, as well as other pharmaceuticals, can upregulate mtDNA replication and maintenance. If successful, this study could identify pharmacological means of mobilization of mitochondrial chaperones to rescue mitochondrial biogenesis in pathogenic conditions.
