Abstract 1116 Fatty Acid Metabolism by CYP168A1: A Cytochrome P450 from Pseudomonas aeruginosa
Pseudomonas aeruginosa is an opportunistic, Gram-negative pathogen that contains three cytochrome P450 enzymes. One P450 found in P. aeruginosa, CYP168A1, is known to be activated in certain disease states and to oxidize fatty acids. With few reaction products identified and its native substrate(s) as-yet unknown, we sought to determine which fatty acids can bind to CYP168A1 and detect the oxidized products made by CYP168A1. After expressing CYP168A1 in E. coli and purifying the protein though IMAC chromatography, we performed ligand binding assays with saturated fatty acids.
